My guess would be that the step and direction wires are either assigned the wrong port and pins, or just wired wrong.

with regards to the uc100, yes that looks right.

I think that part of the Mach3 plugin for the UC100 it allows you to see what the pins are receiving or sending. This will show you what the UC100 is doing.

See part (b) here in the UC100 manual. (note I've never had a UC100).

you can skype, or zoom later if you want and I'll try to help (just PM me) (i've been furloughed so have a little time but am trying to play catchup on hobbies + weather the storm of covid19... in the hope we will all have something to come back to and people still want buildings designing and have money to spend later.... who knows... what else to do but wait and see ... i expect mass layoffs when the government money runs out but who knows....)

These images from the manuals all indicate the same pin numbers to those posted earlier.

PIN14 is noted in the second manual for the controller board as providing the ENABLE but is NOT shown in the Mach3 screenshots.

With the E stop pressed in I am unable to reset Mach3 (normal I assume) and the stepper wheels on the steppers are free to turn by hand.

With the E stop released I am able to reset the Mach3 but once done you hear a click and the hand wheels lock up. Hit the E stop and you hear a click and the hand wheels are released ?????

This is how it should be, it's normal.

Is the Jog on / off button active when trying to move the machine?

Try this too
Try typing in MDI

Well I have good news and bad news......................

The good news is...... its working! 100% fine, jogs no problem, ive even run it through a cycle.

The bad news (well for me anyway) is that it was my fault :? :? :?

It was all due to the motor tuning settings. I was changing them as per the instructions but like an absolute pratt instead of hitting save axis settings I was just hitting OK without even realising there was a button just above saying save axis settings let alone using it. I know, it sounds totally unplausable but here we are. Go easy on me
